Single strand confbrrnation polymorphism (SSCP) is usefuL for the detcction ofpoint mutations in genomic regions and has been applied for mapping known sequence data ofstructurai genes (Hunter et al. 1993, Orita et at. 1989). Two genetic maps have been rcported in L. edodes using RAPD markers (Kwan and Xu 20e2) and AFLP markers (Terashima et al. 2002). These maps were constructed based on the analysis of monokaryotic strains randomly isolated, and the number ot' offgprings used fbr constructing the genetic maps was 32 (Kwan and Xu 2002) and 95 (Terashima et al. 2002). K",an and Xu (2002) employed guantitative trait locus (QTL) analysis to examine the growth rate ot' monokaryotic strains. Agronomic traits have also been characterized by QTL anaLysis in other edibtc basidiomycetes, Agarictts bisporus and Pleurottts ostreatus (Moquet et aL 1999, Larraya et aX. 2002, 2003). The basidiomycete Lentinula edodes (Le.) cytochrome P450, Le.CYP1 was functionally expressed in Saccharomyces cerevisiae. The microsomal fraction containing Le.CYP1 was prepared from the recombinant yeast and the Le.CYP1 was analyzed. The 7-ethoxycoumarin and benzo(a)pyrene were found to be the substrates of Le.CYP1 enzyme. Le.CYP1 converted 7-ethoxycoumarin to 7-hydroxycoumarin.
